Differential expression patterns of cytoskeleton-related genes for meiotic maturation in porcine poor quality and aged oocytes in vitro

During mammalian oocyte maturation, the cytoskeleton plays an important role in spindle assembly. Aduucin α (ADD1) is known to be involved in mitotic spindle assembly as one of the F-actin binding proteins. While many studies examined the arrangement of spindle related to microtubule in oocyte maturation, the loss of F-actin cytoskeleton organization in porcine postovulatory aged oocytes with deteriorated quality during meiosis has not been reported. First, porcine oocytes were divided into two groups (good quality: G1 vs poor quality: G2) based on with/without cumulus cells. The G2 oocytes group caused defects in meiotic maturation capacity (p < 0.05, G1: 82.7 ± 6.3% vs G2: 64.6 ± 5.5%) and impaired (p < 0.05) cytoskeleton organization including spindle assembly and actin dynamics compared to G1 oocytes group. Next, we used aged oocytes cultured for an additional 24 h after in vitro maturation to confirm the changes in cytoskeleton organization in the aged model. Likewise, in aged oocytes, failure of spindle assembly was increased and the expression of F-actin and ADD1 was significantly decreased. In addition, we analyzed specific expression patterns of aging and cytoskeletal-associated genes such as CLU, FAM111A, MAP7, and SKA1 using RNA sequencing analysis. These results indicated that F-actin and actin-binding protein ADD1 play a key role in meiotic maturation via microtubules related to spindle assembly in poor-quality and aged oocytes. Moreover, we suggested diverse aging and cytoskeleton-related genes in porcine oocytes can be effective factors to improve the quality and subsequent developmental capacity by preventing postovulatory aging.
