F-actin interplay coordinates spindle assembly and cytoskeleton organization in porcine poor-quality and aged oocytes during in vitro maturation

While most of the studies have focused on the arrangement of spindle related to microtubule of aging oocyte, the impaired features of oocyte maturation from postovulatory aging such as the loss of F-actin cytoskeleton organization have not been reported. Here, porcine oocytes at germinal vesicle (GV) phase were classified into two groups (good quality: G1 vs poor quality: G2) based on with/without cumulus cells. Meiotic maturation rate derived in G2 group was significantly reduced (p < 0.05, G2: 82.7 ± 6.3% vs G1: 64.6 ± 5.5%) accompanied by DNA damages and loss of cortical F-actin dynamics compared to oocytes of G1 group. Therefore, we also used aged oocytes additional cultured for 24 h after completion of maturation in vitro. We found that loss of F-actin expression in aged oocytes in vitro results in abnormal spindle assembly. F-actin and microtubule cytoskeleton interactions have also been identified as an important feature that supports recovery of oocyte quality and maturation capacity. Additionally, we used RNA sequencing analysis to analyze specific expression patterns of aging and cytoskeletal-associated genes. Our findings showed that F-actin and actin-binding protein adducin are integral components of the meiotic maturation that closely interact with microtubules related to spindle assembly and cortical cytoskeleton structure by using poor-quality and aged oocytes.
